Qualification Details

The candidate is expected to have a PhD degree in bioscience and at least 5-years’ experience from the biotech/pharmaceutical industry and/or from relevant academic research setting. In-depth knowledge and several years of practical experience working with liquid chromatography coupled to mass spectrometry such as e.g. IM-QToF (Waters), Q-Exactive HF Orbitrap (Thermo Scientific) equipped with nano/micro LC, are “a must”. Experience with intact mass determination and protein characterization, including PTM is required.  Detailed understanding of common proteomic assays and techniques, from basic proteolytic digestions to more complex quantitative and qualitative proteomic analysis is desired. Previous experience with characterization and analysis of oligonucleotides is an advantage.

Additionally, hands-on experience with relevant analytical software (such as MassLynx, Unifi, X-calibur, Proteome Discoverer, Compound Discoverer and Trace Finder), analytical sample preparation and reporting of the analysis of test compounds in biological matrices (plasma and tissue homogenates), are crucial.

The successful candidate is expected to have high degree of integrity, motivation, flexibility and capability of working in a structured and efficient way. Excellent interpersonal and cooperation skills as well as ability to work in interdisciplinary teams are essential. Outstanding collaboration skills and dedication to satisfy remote collaborating partner’s needs are expected.

Responsibilities/Job Description

The selected candidate will be responsible for:

  • Intact mass determination of biopharmaceuticals in the range 10-200 kDa using LC-MS.
  • Protein characterization including PTM investigations of antibody drug candidates.
  • Sample preparation and quantitative analysis of in vivo samples of biopharmaceuticals using LC-MS/MS techniques.
  • Develop and execute MS-based assays for proteomic and metabolomic samples, processing and reporting of the results.
  • Ability to develop, evaluate and implement new technologies with focus on expanding the existing LC-IM-HRMS (liquid chromatography-ion mobility -high-resolution mass spectrometry) capacity.
  • Share analytical expertise with other members of the ADMEoT facility and SciLifeLab DDD.
  • Reporting of the results to respective project forums in a timely and orderly manner.

About the Project

Purpose of the position is to provide scientific expertise and contribute to the development of the academic drug discovery (DD) projects ongoing at the SciLifeLab DDD. The major objective is to perform intact mass determination and characterization of biopharmaceutical drug candidates as well as analysis of in vivo (plasma, homogenate target tissues, etc.) samples. The candidate’s responsibilities will include analyzing data, writing reports, and presenting data, both to other DDD facilities and for project meetings. The candidate’s role will also be to complement work of the ADME team with analytical and protein expertise.

About the Department

The Science for Life Laboratory (SciLifeLab) is collaboration between four universities in Stockholm and Uppsala: Stockholm University, Karolinska Institutet, the Royal Institute of Technology, and Uppsala University. The SciLifeLab is a national center for bioscience with a focus on health and environmental research. The center combines advanced technical know-how and state-of-the-art equipment with a broad knowledge in translational medicine and molecular biosciences. As part of this effort, the national platform for academic drug discovery and development was established within SciLifeLab – the SciLifeLab Drug Discovery & Development Platform (SciLifeLab DDD).

ADME of Therapeutics (ADMEoT) facility (Uppsala University Drug Optimization and Pharmaceutical Profiling-UDOPP) is organized at the Department of Pharmacy, Uppsala University and is part of the SciLifeLab DDD. The primary aim of ADMEoT (UDOPP) is to perform ADME (Absorption, Distribution, Metabolism and Excretion), pharmacokinetics (PK) and preclinical characterization of hit collections and compound series during the lead generation/ optimization stage of the drug discovery process. The facility is working with small molecules and proteins i.e. antibodies drug candidates.
